Bmax -- Maximal Binding (fmol/mg protein), Kd -- dissociation constant (nM). Values are Mean ± S.E.M. of 4-6 separate experiments. Each group consists of 6-8 rats. a p < 0.001, b p < 0.05 when compared to control group; c p < 0.001 when compared to diabetic group. Incubation was done with 1.0 nM-30 nM at 37 °C of [3H] 5-HT in a total incubation volume of 250 μl. 10 μM unlabelled 5-HT was used to determine the nonspecific binding. The reaction was stopped by rapid filtration through GF/B filters using ice cold Washing Buffer pH 8.5. Bound radioactivity was counted with cocktail-T in a Wallac 1409 liquid scintillation counter.
