In vivo tumor treatment experiments. C57BL/6 mice (5 per group) were challenged subcutaneously with 1×104/mouse of TC-1 tumor cells. Three days later, the mice were immunized subcutaneously using HPV-16 E7 (aa 49-57) peptide with PADRE peptide or with poly(I:C) or with both poly(I:C) and PADRE peptide. Tumor-bearing mice treated with PBS or with poly(I:C) and PADRE without E7 peptide were used as controls. The mice were given booster with the same dose every week at the same site until they died or the tumor reaches 2 cm in diameter. The survival of tumor-bearing mice was analyzed by Kaplan & Meier analysis.
