Effect of different extracellular calcium concentrations ([Ca2+] o ), the CaSR antagonist (NPS2390), blocker of L-type calcium channels (CdCl 2 ), and inhibitor of Na+-Ca2+exchanger (NiCl 2 ) on the intracellular calcium concentrations ([Ca2+] i ) in the PAMSCs. A. [Ca2+]o from 5 to 12.5 mM caused an increase fluorescent intensities of the [Ca2+]i in a concentration dependent manner, then we chose 10 mM [Ca2+] for the futher experiments (n = 20). B. The cells were exposed to 10 mM Ca2+, and FI of [Ca2+]i was recorded for 120 s. In some experiments, the cells were pre-exposed to 0.1 mM NiCl2, 0.02 mM CdCl2, and 10 μM NPS2390 for 30 min before Ca2+ challenge.
