O -GlcNAcylation of Thr-366 and Thr-367 is involved in KSHV reactivation. (A) Comparison by flow cytometric analysis of the potency for viral latency disruption of K-RTA and O-GlcNAc mutants. 293/rKSHV.219 cells transfected with K-RTA or the three O-GlcNAc mutants (T366A, T367A and T366A/T367A) for 72 h were analyzed on a fluorescence-activated cell sorter followed by scoring the percentage of double positive (GFP and RFP) cells in each population, as indicated in the upper right of each panel. Untreated cells (293/KSHV) served as a control. (B) A schematic chart summarizes the results derived from eight independent experiments.
