AACOCF3 and MAFP eliminated H-rev107-mediated H-RAS palmitoylation. HtTA cells plated in a 10-cm dish were transfected for 24 h with 0.1 μg of H-RAS along with H-rev107-myc or the control vector in the presence of 10 μM AACOCF3 (A, B), 1 μM palmostatin B (A), 5 μM MAFP (B), 2 μM BEL (B) or DMSO vehicle. Cell lysates were prepared, and acyl-biotin exchange analysis of H-RAS was performed as described in Methods. Levels of H-RAS and H-rev107 in the cell lysates are shown in the bottom panel. NH2OH: hydroxylamine.
