Effect of CAPE on the SMase/ceramide pathway. (A) Time course showing the effect of CAPE on intracellular SMase activation in C6 glioma cells. Cells were treated with CAPE (50 μM) for the indicated times, and SMase activation was analyzed using a fluorometric assay in which SMase activity was directly proportional to the fluorescence emitted. Data are presented as the mean ± S.D. of three independent experiments. (B) Time course showing the effect of CAPE on intracellular ceramide levels in C6 glioma cells. Cells were treated with CAPE (50 μM) for the indicated times, and ceramide was resolved by HPTLC. Std, standard (C-2 ceramide).
