From: SB203580 increases G-CSF production via a stem-loop destabilizing element in the 3’ untranslated region in macrophages independently of its effect on p38 MAPK activity

SB203580 slows G-CSF mRNA degradation in RAW264.7 macrophages. a RAW264.7 cells were pretreated for 30 min with DMSO or 10 μM SB203580, then 100 ng/ml of LPS was added and incubation continued for 5 h, then the cells were treated with actinomycin D (4 μg/ml) for 0, 1, 2, or 3 h. The G-CSF mRNA levels were determined by RT-qPCR, normalized to GAPDH mRNA levels, and expressed relative to the value at time 0, set as 100 %. The results are the mean ± SD for three independent experiments. *p < 0.05, compared to cells not treated with SB203580 at the corresponding time point. b RAW264.7 cells were pretreated for 30 min with DMSO or 10 μM SB203580, then actinomycin D (4 μg/ml) was added for 0, 0.5, 1, 2 or 3 h. Then the G-CSF mRNA levels were determined by RT-qPCR and normalized to GAPDH mRNA levels and expressed relative to the value at time 0 (relative value =100 %). The results are the mean for two independent experiments
