No significant differences in the efficiency of transgene integration between different lengths of overhangs. HEK293T cells were transfected with plasmids expressing either Cas9 D10A nickases or H840A nickases, alongside plasmid containing AAVS1 pair guides and the donor plasmid. Cells were analysed for GFP expression by flow cytometry 2 days after transfections. A Strategies utilizing 5ʹ overhang DNA DSBs with various lengths of overhangs; B strategies utilizing 5ʹ overhang DNA DSBs with various lengths of overhangs.
